A short-acting GHRH analog (Modified GRF 1-29) without the Drug Affinity Complex, producing brief, pulsatile growth hormone release that more closely mimics natural GH secretion patterns.
Modified GRF 1-29 (also called CJC-1295 without DAC or Mod GRF 1-29) is a synthetic analog of the first 29 amino acids of growth hormone-releasing hormone with four amino acid substitutions (Ala2, Gln8, Ala15, Leu27) that confer resistance to enzymatic degradation by dipeptidyl peptidase IV (DPP-IV). Without the DAC moiety, its half-life is approximately 30 minutes, producing a transient pulse of GH release from anterior pituitary somatotrophs. This short-acting profile better mimics the natural pulsatile pattern of GH secretion and is often combined with growth hormone-releasing peptides (GHRPs) for synergistic effects.

Key finding: Review of GHRH(1-29) analogs examining the pharmacology of pulsatile GHRH receptor activation for GH release, establishing the foundational receptor pharmacology shared by Mod GRF 1-29.

Key finding: Measured GHRH(1-29)-NH2 pharmacokinetics and the growth hormone response it produced in healthy subjects across two routes of administration.
